Is it more accurate to tune your engine off the ground?

No. You need a load on it to tune it correctly. A good sounding engine on the stand will usually be too lean when it makes contact with the ground and is loaded. You can get it maybe close and leave it a little rich sounding, but final tune is when you are on the ground loaded.
